abstract | (57) [Summary] [Object] To provide a method for producing and assaying hybrid cells of Strophanthus genus useful as a cardiac glycoside-producing plant. [Structure] Strophanthus glatus cells, which are milky white under bright culture conditions, and Strophanthus amboensis cells, which are yellow-green under bright culture conditions, are used as protoplasts, and both protoplasts are fused by electrofusion. . This is cultivated in a medium in which protoplasts of Gratas can grow but protoplasts of Amboensis do not, and callus is grown from the obtained colonies, and those which become yellow-green callus are selected as hybrid cells. The assay of hybrid cells is carried out by using the cells provided for the production of hybrid cells and the DNA of these hybrid cells as a restriction enzyme Nhe. It is digested with I, separated by electrophoresis, and hybridized by using the ribosomal RNA gene of rice as a probe. |
