(57) [Summary] [Objective] Increased expression level of structural gene encoding a polypeptide having HMG-CoA reductase activity in mutant yeast having single or double deficiency in sterol biosynthetic enzyme expression. To increase the accumulation of squalene and specific sterols in yeast. [Structure] The expression level of the structural gene is adjusted so that a foreign DNA fragment encoding a polypeptide having HMG-CoA reductase activity and a promoter suitable for directing the expression of the encoded polypeptide in transformed yeast. It is desirable to elevate by transforming yeast with a recombinant DNA molecule containing a vector operably linked to. The polypeptide having HMG-CoA reductase activity is preferably a truncated, active HMG-CoA reductase. Also disclosed are recombinant DNA molecules useful for transforming yeast, and yeast transformed with such DNA molecules. |
