titleOfInvention Method for transforming Lactobacillus of Lactobacillus and transformant thereof
abstract (57) [Summary] [Object] The present invention provides a method for transforming Lactobacillus of the genus Lactobacillus and a transformant thereof. [Structure] The main point is that the cells can be replicated by the bacterium after the cells have been subjected to a temperature treatment of, for example, 45 ° C. for 30 minutes using cells that have been cultivated in a medium in which the pH has been lowered and formate has been added. An electric pulse is applied together with such a plasmid (electroporation method), the cells are expressed and cultured in a medium suitable for the bacterium, and then cultured in a selective medium to obtain a transformant. [Effect] Lactobacillus Lactobacillus del brückii subsp. bulgaricus Is a lactic acid bacterium that is industrially important in the production of yogurt and the like. However, despite many studies, transformation with this bacterium has not been successful so far, and the success by the method of the present invention is the first example. Using this method, different types of messages (Strept ococcus salivarus subsp. Since the L-lactate dehydrogenase gene derived from thermophilus) was successfully introduced into this bacterium and expressed, the utility of the present invention was proved.
