titleOfInvention Method for measuring DNA or RNA fragment using immobilized complementary DNA or RNA
abstract (57) [Summary] [Objective] To improve the measurement sensitivity and reliability in the method of measuring DNA or RNA fragments by the fluorescence polarization method. [Structure] (1) Fluorescently labeled single-stranded DNA or RNA probe and (2) DNA or RNA in a sample (3) competitive reaction with an immobilization reagent in which DNA or RNA containing a base sequence complementary to the single-stranded DNA or RNA is bound to an immobilization carrier to give double-stranded DNA or R DNA in the sample is formed by forming NA and measuring the displacement between the fluorescence polarization degree before formation of the double strand and the fluorescence polarization degree after formation of the double strand. Alternatively, the base sequence corresponding to the single-stranded DNA or RNA probe existing in RNA is measured. [Effect] Immobilized DNA or RN containing a fluorescently labeled DNA or RNA and a base sequence complementary to the measurement target Since the effective molecular weight change due to the complementary binding reaction with A is large, the change in the fluorescence polarization degree is large, and the measurement sensitivity and reliability are improved.
