abstract PURPOSE: To detect the subject virus for diagnosis of C-type hepatitis in high sensitivity even in an initial stage of affection by synthesizing a cDNA from a nucleic acid prepared from a test substance and amplifying the cDNA polymerase using a specific oligo-DNA set as a primer. n CONSTITUTION: Blood serum of a patient diagnosed as non-A, non-B type chronic hepatitis is ultracentrifuged for 5hr and a resultant pellet is suspended in a buffer solution to prepare a virus suspension, then the suspension is treated with adding proteinase in the presence of a surfactant, thus extracted with phenol/chloroform and an RNA is precipitated with ethanol, then a cDNA is synthesized using said RNA as a mold, thus the cDNA is amplified in treating with DNA polymerase to obtain a DNA group 1 expressed by formula I to formula VI and a DNA group 2 expressed by formula VII to formula XIII. Next, a cDNA is synthesized from a nucleic acid prepared from a test substance and the cDNA is amplified using DNA polymerase and an oligo-DNA set respectively at least a species selected from the DNA group 1 and the DNA group 2 as primers to detect C-type hepatitis virus in high sensitivity. n COPYRIGHT: (C)1992,JPO&Japio
