PURPOSE: To provide a new antibody-nonproductive cell strain to be used for producing IgM or IgA antibody containing human J-chain protein. n CONSTITUTION: The objective antibody-nonproductive cell strain, which does not produce endogenous antibody and H-chain and L-chain as its constituents, obtained by introducing exogenous antibody gene into namaruba cell strain assimilated to a serumless medium. Specifically, antibody-nonproductive mutant cell strain is first obtained, by X-ray irradiation or using a mutating chemical, from the namaruba cells (ATCC CRL 1432) of human B lymphocyte strain, pref. NKG4 strain, and said mutant cell strain is put to cloning using a serumless medium to enable said strain to be cultured in such serumless medium, thus obtaining the objective antibody-nonproductive cell strain sensitive to amidoglycoside-based antibiotic G-418 or mycophenolic acid. n COPYRIGHT: (C)1992,JPO&Japio |
