abstract | PURPOSE: To purify a high-purity DNA in high yield by simple operation without using a harmful reagent by subjecting cells of bacterium to bacteriolysis, passing the cells through a membrane filter to remove insoluble substances, removing impurities by ultrafiltration and concentrating the DNA. n CONSTITUTION: Escherichia coli transformed with a recombinant plasmid (e.g. plasmid pUC119) is cultured in an ampicillin-containing medium at 37°C for 24 hours, a culture solution is centrifuged to collect cells, which are suspended in a tris-hydrochloric acid buffer solution (pH 8) containing 10mMEDTA, subjected to bacteriolysis with 0.2N NaOH-1% sodium dodecyl sulfate, then the mixed solution is filtered under pressure by a membrane filter having 0.1-2μm pore diameter to remove insoluble substances, subjected to ultrafiltration with an ultrafilter having 30,000-1,000,000 fraction molecular weight to remove impurities and concentrated to separate and purify plasmid and/or cosmid DNA from cells of bacterium. n COPYRIGHT: (C)1992,JPO&Japio |
