abstract | PURPOSE: To obtain the title compound in a state of crystal useful as a synthetic intermediate for 1β-methyl-carbapenem-based antibiotics, having excellent solid stability. n CONSTITUTION: A (1R,5R,6S)-p-nitrobenzyl-2-(diphenylphosphorinoxy)-6-[(R)-1- hydroxyethyl]-1-methyl-carbapenem-3-carboxylate shown by formula I (Ph is phenyl; RNB is p-nitrobenzyl) in a state of crystal. The crystal, for example, is obtained by reacting p-nitrobenzyl(1R,5R,6S)-[(R)-1-hydroxyethyl]-2-oxo- carbapenem-3-carboxylate shown by formula II with a diphenyl phosphoric acid halide in an inert solvent such as DMF in the presence of a base such as triethylamine, concentrating the reaction solution, then purifying the residue by silica gel column chromatography to give powder, dissolving the powder in ethyl acetate under heating and successively cooling at room temperature. n COPYRIGHT: (C)1992,JPO&Japio |
