abstract PURPOSE: To provide a new peptide derivative which is a kind of vasoactive intestinal peptide having a specific amino acid sequence, exhibiting strong blood flow increasing action and low hypotensive side effect and effective for ulcer, chilblain, impotency, hair-growth, etc. n CONSTITUTION: A protected peptide-MBHA resin is synthesized by successively bonding an amino acid having protected α-amino group and side-chain functional group from C-terminal side according to the amino acid sequence of the objective peptide using an MBHA resin as a solid phase carrier in a solid-phase synthesis using a peptide synthesizer and repeating the above steps. The synthesized resin is treated with anhydrous hydrogen fluoride to eliminate the peptide from the resin and remove all protection groups from the peptide. After distilling out the anhydrous hydrogen fluoride under reduced pressure, the residue is added with 0.1M acetic acid to extract the peptide and the extract liquid is purified by ion-exchange column chromatography followed by high-performance liquid chromatography to obtain the objective peptide of formula (A, C and D are Lys or Arg; B is Met, etc.). n COPYRIGHT: (C)1992,JPO&Japio
