abstract | PURPOSE: To provide a novel labeling reagent that enables in situ detection of a chromosome or a region of a chromosome. n CONSTITUTION: Phenyl multiple labels are covalently linked to one or more DNAs complementary to the nucleic acid sequence to be detected through amino acid-linking group (lower alkyl amino acid or their polymer or copolymer). The DNA sequence before labeling has about 300 bp length on the average and is obtained from the recombinant phage library of the chromosome or a region of the chromosome to be detected. The phenyl labels are 4-nitrophenyl, 2,4-dinitrophenyl or 2,4,6-trinitrophenyl. In order to form covalent bonds, amino acidlinking group is allowed to react with a label of phenyl group bearing 1-3 nitro groups. The resultant label-linker compound is activated by esterification to 3-sulfo-N-hydroxysuccinimide and the activated label-linker compound is covalently linked to the amino group-rearranged DNA. n COPYRIGHT: (C)1992,JPO |
