abstract | PURPOSE: To provide a new steroid derivative composed of 3-oxo-1,4-pregnadiene derivative, free from glucocorticoid action, having highly selective strong local antiinflammatory action and useful as antiinflammatory agent having low side effect. n CONSTITUTION: The objective steroid derivative of formula III [X is H or F; R 1 is H, OH or methyl; R 2 and R 3 are H, OH, O(CH 2 ) n SiR 3 (R is alkyl; n is 0 or 1-6), etc.; R 4 is alkyl] [e.g. methyl(20S)-acetoxy-11β,17-dihydroxy-3-oxo-1,4- pregnadien-21-oate] can be produced by treating a steroid having hydroxymethyl ketone group at 17-site and expressed by formula I (e.g. prednisolone) in anhydrous methanol in the presence of anhydrous copper acetate at ordinary temperature to obtain a trihydroxy compound of formula II (methyl 20-11β,17,20- trihydroxy-3-oxo-1,4-pregnadien-21-oate) and acylating, alkylating or silylating the 20-OH group of the product. n COPYRIGHT: (C)1992,JPO&Japio |
