abstract | PURPOSE: To selectively acylate the 32-position of the subject compound without using toxic pyridine by bringing an FK-506 type macrolide into contact with an acyl donor and a lipase enzyme immobilized on a support in an organic solvent for a long time. n CONSTITUTION: A lipase N enzymic solution is made to act on a solid support such as Celite in a phosphoric acid buffer, then dried at 40°C under reduced pressure with a rotary evaporator to afford an immobilized lipase enzyme. An FK-506 type immunosuppressant macrolide such as immunomycin (FK-520) is then added thereto under conditions for using an acyl donor (e.g. acetic anhydride) and an anhydrous organic solvent without hydroxyl group (e.g. benzene) and brought into with contact with the immobilized lipase enzyme under reactional conditions for a sufficiently long time to selectively acylate the C-32- position.
