abstract | PURPOSE: To readily measure the subject substance effective in diagnosis, etc., of hypercalcemia caused by malignant tumor by reacting a specimen with an antibody specific for a parathyroid hormone-related protein and detecting the produced antigen-antibody reaction. n CONSTITUTION: A parathyroid hormone-related protein (PTHrP) expressed by the formula is reacted with Na 125 I using chloramine T to prepare a 125 I- labeled PTHrP. A prescribed amount thereof is then mixed with a specimen such as blood of a patient and the resultant mixture of the antigen with the labeled antigen is subsequently reacted with an antibody specifically recognizing C-terminals of the PTHrP. The produced antigen-antibody complex is then precipitated by centrifugation and separated from the unreacted antigen and antibody. The radiation dose of the formed precipitates is measured with a well type scintillation counter. The measured value is subsequently used to read the concentration from a standard curve prepared by measuring an antigenic standard reagent at well-known concentrations according to a similar method.
