titleOfInvention Production (r)-2-hydroxy-4-phenylbutyric acid
abstract PURPOSE: To produce the subject compound in high purity and efficiency by reacting racemic 2-hydroxy-4-phenylbutyric acid (HPB) with a carboxylic acid anhydride, etc., in the presence of a specific enzyme. n CONSTITUTION: Racemic HPB (A) is charged in an amount of 0.5-50wt.% to an organic solvent such as toluene to obtain a reaction liquid (B). A carboxylic acid anhydride such as acetic anhydride is added to the liquid B in an amount of 0.5-10 equivalent based on the component A. A reaction mixture (C) is produced by adding 0.1-5 pts.wt. (based on 1 pt.wt. of the component A) of a lipase produced by a microorganism belonging to genus Pseudomonas. The component C is made to react at 5-50°C to effect the acylation of (S)-HPB and obtain a reaction product (D). The objective (R)-2-hydroxy-4-phenylbutyric acid is produced by filtering the component D to remove insoluble materials, distilling out the reaction solvent and purifying the residue by column chromatography, etc. n COPYRIGHT: (C)1992,JPO&Japio
