abstract PURPOSE: To enable creation of new Acremonium.chrysogenum by utilizing a DNA compound capable of coding cephalosporin C acetyl esterase derived from the Acremonium.chrysogenum. n CONSTITUTION: A DNA is extracted from a selected phage and southern hybridization is carried out by using a probe to position the objective gene on a restriction enzyme fragment of a suitable size. The obtained hybrid is then subcloned into a plasmid vector. The base sequence of the resultant DNA fragment is subsequently determined to confirm the presence of a cephalosporin C acetyl esterase genomic DNA by comparison with the well-known amino acid sequence. A DNA fragment of a suitable size containing the whole coding region of the cephalosporin C acetyl esterase is integrated into a vector for transforming the Acremonium.chrysogenum to prepare a recombinant DNA, which is then used to transform the Acremonium.chrysogenum. n COPYRIGHT: (C)1992,JPO&Japio
