PURPOSE: To produce serum albumin free from the problem of viral infection by culturing globular cultured hepatocyte collected from fresh liver of matured animal and separating the albumin from the cell and/or the culture medium in purified state. n CONSTITUTION: Flawless hepatocyte collected from fresh liver of a matured animal is cultured in a serumfree medium produced by adding insulin, glucagon, epidermal growth factor, prolactin, growth hormone, copper and zinc to William' s E medium. Since the hepatocytes form a floating cluster consisting of cells agglomerated in globular form, the globular cultured hepatocytes are cultured. The objective serum albumin is separated in purified state from the cell and/or the culture medium by conventional method such as gel-filtration, low- temperature ethanol precipitation and affinity chromatography. A serum albumin free from the contamination of virus, etc., can be produced by this process in high yield without using serum. n COPYRIGHT: (C)1991,JPO&Japio |
