abstract | PURPOSE: To amplify a gene for detecting a specific sequence, by bringing two primer nucleic acids being immobilized or immobilizable into contact with a test sample and treating the resultant product with an extender enzyme and a nucleotide residue. n CONSTITUTION: Two primer nucleic acids in which one nucleic acid is being immobilized or immobilizable and the other is being immobilized, immobilizable or labeled in a nonisotopic way are brought into contact with a test sample containing a target nucleic acid sequence under hybridization conditions. The resultant product is treated with an extender enzyme and at least one nucleotide residue (preferably nucleotide triphosphate) and the resultant hybridized primer nucleic acid is elongated in the direction opposite to a solid substance to produce a target nucleic acid sequence immobilized by denaturing.
