abstract | PURPOSE: To allow erythrocytes to be sufficiently solved while maintaining their light scattering characteristics for a time required for classifying subgroups of leukocytes, by aqueous solution comprising aromatic oxyethanol, specific organic buffer agent and nonionized surfactant. CONSTITUTION: Solving agent comprises aromatic oxyethanol, organic buffer agent serving to provide pH buffer ability and increase conductivity having pH of 8.5 or its vicinity and nonionized surfactant components. The aromatic oxyethanol is preferably 2-phenoxyethanol, which solves erythrocytes and coats leukocytes, and also serves as antibacterial agent. After the agent is added to diluted full blood sample, a converged laser beam is intersected with the erythrocytes and leukocytes, then four light scattering parameters are measured at 0 deg., 10 deg., 90 deg. and depolarized 90 deg., and these are analyzed to obtain five subgroup classification counts. Thus, leukocytes can be classified into groups, wherein light scattering characteristics can be maintained for a time required for the classification, thereby allowing erythrocytes to be sufficiently solved. |
