titleOfInvention Production of optically active 3-aryl-3-hydroxypropionic acid alkyl ester
abstract PURPOSE: To produce a large amount of the subject compound in high purity and efficiency by treating a 3-aryl-3-hydroxypropionic acid alkyl ester (AHPA) with an esterase originated from microorganisms, etc. n CONSTITUTION: A reaction liquid is produced by mixing a substrate consisting of an AHPA of formula I (R 1 to R 5 are H, OH, 1-4C alkoxy or benzyloxy; R 6 is alkyl) and a fatty acid vinyl ester or triglyceride at a molar ratio of 1:(0.5-10) and dispersing the mixture in a solvent such as n-hexane. An esterase originated from microorganisms of genus Pseudomonas, etc., is added to the reaction liquid and reacted at 10-150°C for 1-1,000hr to effect the ester interchange with the triglyceride to obtain an optically resolved product. The product is filtered, distilled under reduced pressure and subjected to a column chromatography to separate an optically active 3-aryl-3-acyloxypropionic acid alkyl ester and produce an optically active 3-aryl-3-hydroxypropionic acid alkyl ester of formula II [e.g. (-)-3-(2-methoxyphenyl)-3-hydroxypropionic acid methyl ester]. n COPYRIGHT: (C)1991,JPO&Japio
