abstract | PURPOSE: To synthesize human-monocyte-CSF growing agent by culturing human recombination type M-CSF prepared from human urine derived macrophage colony stimulating factor (M-CSF) and M-CSF gene. n CONSTITUTION: Human derived M-CSF and M-CSF gene having about 4Kb are transduced to CHO cells, etc., by using calcium phosphate to produce human recombination type M-CSF. The human recombination type M-CSF is adjusted to about 1×10 3 to 1×10 5 human monocytes/ml concentration by using preferably RPMI 1,640 medium and adding about 5-20% bovine fetal serum, put in a laboratory dish of plastic and cultured. The medium is changed to a new medium containing M-CSF at intervals of about three days and after 2-4 weeks, multiplied human monocyte-macrophage is released and recovered by using rubber policeman, etc. n COPYRIGHT: (C)1991,JPO&Japio |
