abstract | PURPOSE: To enable the separation of high density lipo proteins from the whole blood by mixing a whole blood sample subjected to an anti-coagulation processing to precipitate low density lip proteins, super low density lipo proteins and chylomicron for centrifugal separation. CONSTITUTION: The whole blood is mixed with an aqueous solution which comprises magnesium ++ cation and dextrane sulfate 500 at a ratio and at a concentration that allow low density lipo proteins(LDL), super low density lipo proteins(VLDL) and chylomicron to be precipitated while high density lipo proteins (HDL) are left in a solution. Then, the lipo proteins, chylomicron and red blood cells precipitated are settled by centrifugal separation exceeding 1,000xg. Then, a recovery of HDL is performed at a plasma density of 1,210g/ml. Then, the fraction of the isolated lipo proteins is quantitatively recovered and the contents of cholesterol, triglyceride, lipophosphate or proteins are measured, thereby enabling determination. |
