New arginine deiminase, production thereof and anticancer agent containing same enzyme as active ingredient |
abstract |
NEW MATERIAL:An arginine deiminase having the following physicochemical properties: Action thereof and specific property of substrate, capable of producing L-citrulline and ammonia by hydrolyzing amidino group of L-arginine; optimum pH, 6.0-7.5; stable pH, 4.5-9.0; optimum temperature, about 50°C; Km value, 0.2mM; isoelectric point (pI), 7; molecular weight, about 45000 (SDS- polyacrylamide gel electrophoresis method) or about 90000 (gel permeation HPLC method); amino acid sequence from N end, sequence expressed by the formula. n USE: Anticancer agent. n PREPARATION: Mycoplasma [preferably Mycoplasma arginine (IFO No. 14476)] having arginine decomposing ability is cultured and cultured material of the germ cell is extracted and collected by separating and purifying from the extract. n COPYRIGHT: (C)1991,JPO&Japio |
