abstract | PURPOSE: To enable diagnosis at a high accuracy by combining a specific antibody with a monoclonal antibody capable of inhibiting activity of salivary-α-amylase. n CONSTITUTION: Cell capable of producing an antibody of an animal immunized with salivary-α-amylase (A) as an antigen and animal myelomatous cells are fused to provide fused cells, which are then cultured to afford the first monoclonal antibody (B). An immune complex of the ingredients (A) and (B) as an immunogen is administered to an animal to afford fused cells, which are subsequently cultured to provide the second monoclonal antibody (C) having the following properties. Belonging to IgG 1 subclass. Precipitating with 50% saturated (NH 4 ) 2 SO 4 solution and binding to protein A by dialyzing the resultant precipitates with a physiological saline solution, then mixing thereof with an equal amount of 1.5M glycine-3.0M NaCL (pH9) solution. Eluting with 0.1M citric acid at pH3 by protein A column chromatography. Inhibiting ≤85% activity of the ingredient (A) alone and ≥99% activity of the ingredient (A) by combination with the ingredient (B), etc. The ingredients (B) and (C) are subsequently combined to fractionate and determine pancreatic-α-amylase. n COPYRIGHT: (C)1991,JPO&Japio |
