abstract PURPOSE: To obtain the subject new enzyme having various prescribed physicochemical properties and capable of production of a specified optically active butyric acid derivative using an enzymatic reaction thereof from the cultured material obtained below by culturing a microorganism belonging to Leuconostoc genus or Lactobacillus genus. n CONSTITUTION: The above-mentioned microorganism (preferably Leuconostoc mesenteroides subsp. dextranicum IFD 3349, etc.) is cultured in a culture medium capable of growth of the above-mentioned microorganism. As the culture method, the culture is carried out anaerobically or aerobically in a culture medium having pH3.0-9.5, preferably 4-8, at 20-45°C, preferably 25-37°C, for about 5-120hrs. The resultant cultured material is subjected to, e.g. centrifugal separation to recover the cell bodies and the recovered cell bodies are subjected to, e.g. ultrasonic crushing to obtain an cell-free extract. The resultant extract is then purified by streptomycin sulfate treatment, ammonium sulfate fractionation, the ion-exchange chromatography, etc. The obtained enzyme shows physicochemical properties represented by (1)-(8) and (R)-2-hydroxy-4-phenylbutyric acid can be produced by an enzymatic reaction thereof. n COPYRIGHT: (C)1991,JPO&Japio
