titleOfInvention | Retinoid receptor |
abstract | PURPOSE: To measure retinoic acid with a good sensitivity by using the retinoid receptor prepd. from the intestinal canals (including the duodenal tubes) of fowls. n CONSTITUTION: The embryos of fowl, such as chickens and ducks or the intestinal canals of chicks right after hatching are enucleated and finely segmented and are then subjected to a homogenization treatment in a buffer soln. to obtain the coarse extract of the retinoid receptor. This extract is refined by a centrifugal sepn. and is disposed into a container where the extract is freeze-dried. The inside of the container is substd. with an inert gas after drying and the container is hermetically closed to obtain the prepn. of the retinoid receptor. The characteristics of the retinoid receptor has the strong affinity to retinoid and the bond constant thereof exhibits order of 10 -9 to 10 -11 M. Order of the specificness of Ch 55, retinoic acid and Am 80 is Ch 55>retinoic acid>Am 80 and the apparent mol. wt. exhibits 0 to 35 kilo dalton with a monomer and 75 to 85 kilo dalton with a dimer. Further, the retinoid receptor is stabilized in the copresence of an SH group reducing agent (e.g.: dithiothreitol) and/or retinoid. n COPYRIGHT: (C)1991,JPO&Japio |
