abstract | PURPOSE: To obtain a nucleoside phosphorylase exhibiting specific physical and chemical properties and having the activities of purine as well as pyrimidine nucleoside phosphorylase. n CONSTITUTION: The nucleoside phosphorylase has the following physical and chemical properties I to VI. (I) Action: catalyzing the reaction of formula 1 (B is purine base or pyrimidine base; R is OH or H). (II) Substrate specificity: acting to purine nucleoside and 2'-deoxypurine nucleoside excluding xanthosine and acting to pyrimidine nucleoside and 2'-deoxypyrimidine nucleoside. (III) Optimum pH is about 8.0 and the compound is stable between pH6.5-9.0 by the treatment at 4°C for 24hr. (IV) The optimum temperature is about 40°C and the compound is stable up to 60°C by the treatment at pH7.0 for 30min. (V) Molecular weight: about 125,000 by gel-filtration and about 25,000 by sodium dodecyllaurylsulfate-polyacrylamide gel electrophoresis and the molecule is constituted of 5 subunits.
