abstract | PURPOSE: To perform measurement of activity of enzyme peroxide with high sensitivity by using H 2 O 2 , 3-methyl-2-benzothiazolinone hydrazonate (MBTH) and arylalcohol derivative and measuring said activity in pH range near neutral. n CONSTITUTION: A solution containing enzyme peroxide or a solid phase containing enzyme peroxide in enzyme immunoassay is reacted with a solution containing H 2 O 2 , MBTH and arylalcohol derivative and activity of said enzyme is measured. As an essential condition, said measurement is performed within a range of pH6.9-7.2. MBTH may be hydrochloric acid or other water-soluble salt such as sulfate and a compound expressed by formula I or II is used as arylalcohol derivative. In the measurement, phosphoric acid buffer solution is preferably used, reaction temperature is usually in a range of room temperature - 37°C, dimethylsulfoxide, etc., may be used as reaction stopping solution and absorbance may be measured by a spectrophotometer using a reaction buffer solution not containing enzyme peroxide as a reference after stopping of the reaction. n COPYRIGHT: (C)1990,JPO&Japio |
