PURPOSE: To obtain an enol-ether type precursor useful for the synthesis of 1,2-dioxetane used for testing based on chemiluminescent decomposition by o-alkylating a cycloalkyl aryl ketone. n CONSTITUTION: A compound of formula I (T is polycycloalkyl; R 1 is 1-20C alkyl, aralkyl, cycloalkyl; Y is fluorescent chromophore), for example, 3- methoxyphenyl adamant-2-yl ketone is reacted with an alkylating agent such as dimethyl sulfate, in a polar non-protonic solvent in the presence of an alkali metal alkoxide to obtain a compound of formula II (R is R 1 ), for example, (3-methoxyphenyl)methylene adamantane. Dioxetane is used for an antigen- antibody reaction, an enzyme immunoassay, and a polynucleotide probe method. n COPYRIGHT: (C)1990,JPO |
