abstract | PURPOSE: To readily and speedily determine theophylline by reacting an alkali phosphatase with a substrate in the presence of a buffer and polyvinylpyrrolidone(PVP) present in a specific subbing zone. n CONSTITUTION: (a) A porous spreading layer is obtained by allowing ≥1 g/m 2 buffer such as tris(hydroxymethyl)aminoethane hydrochloride and 1-5 g/m 2 substrate such as p-nitrophenyl phosphate to be impregnated into a fibrous material. (b) A subbing layer is obtained by mixing PVP and a buffer of pH≤9. (c) A reagent layer is obtained by adding ≥10 I.U./m 2 APH acting on a substrate at pH≤9. The objective analytical element for theophylline determination is obtained by individually supporting the components (a) to (c) and (d) a radiation- blocking layer on (e) a supporter such as the one of PE. The amount of theophylline is determined by bringing the element into contact with a fluid sample of a living body such as a human being, heating the element keeping contact with the fluid sample to react, developing the color of not inhibited APH corresponding to the amount of the theophylline in the sample, and measuring the developed color by a spectrophotometer, etc. n COPYRIGHT: (C)1990,JPO |
