Production of 4-alkoxyalkyl-4-aminophenylpiperidine and its derivative |
abstract |
PURPOSE: To obtain the subject compound useful as an analgesic for intravenous injection or an anesthetic by reacting an N-substituted 4-piperidone compound with an anionic reagent and reducing the product with a reducing agent. n CONSTITUTION: An N-substituted 4-piperidone compound is reacted with an aniline compound to form a Schiff base compound, and this is reacted with an anion-stabilizing group-containing anionic reagent represented by the formula [wherein X is an anion stabilizing group; Y is H or a lower alkyl; Z is a lower alkoxyl or phenylmethoxyl; and M is an alkali or (alkaline earth) metal] to obtain an amine compound. This is reduced with a reducing agent (e.g. metal hydride or Raney nickel) to stabilize the anion stabilizing group to obtain a 4-alkoxyalkyl-4-aminophenylpiperidine. Suitable examples of the anionic reagents include methoxymethylphenyl sulfide and an α-Li derivative thereof, etc. n COPYRIGHT: (C)1990,JPO |
