abstract PURPOSE: To obtain a monoclonal antibody by fusing an antibody-forming cell of mammals except human immunized against β chain manifesting cell strain YTS with a cell having permanently multiplying properties to give a hybridoma and culturing the hybridoma. n CONSTITUTION: An antibody-forming cell of mammals except human immunized against β chain manifesting cell strain YTS is blended with a cell having permanently multiplying properties and subjected to cell fusion by gradually adding a fusion promoter such as polyethylene glycol while maintaining the reaction mixture at about 37°C. Then the fusion promoter added to the culture solution is diluted to stop the fusion and the cell is separated by centrifugal separation. The cell is cultured in a hypoxanthine aminopterin thymidine to give a hybridoma. The prepared hybridoma is cultured in vitro or in vivo to produce a monoclonal antibody. n COPYRIGHT: (C)1990,JPO&Japio
