abstract | PURPOSE: To obtain a preparation containing an effective amount of a stabilized polyol and a sufficient amount of a buffering agent to maintain the pH level within a specific range, with physical stability, chemical stability and biological activity maintained, and capable of controlledly releasing somatotropin over a long period. n CONSTITUTION: This somatotropin preparation contains (A) ≥10 (pref. ≥20)wt.% of biologically active somatotropin, (B) an effective amount (20-80wt.%) of a polyol such as glycerol or tris(hydroxymethyl)aminomethane, and (C) a sufficient amount of a buffering agent (e.g. histidine hydrochloride, alkali metal phosphate) to attain the pH level so as to maintain the biological activity of the somatotropin over a sufficiently long period (namely, between 4.5 and a greater value, either 7 or the isoelectric point of the somatotropin). It is preferable that a wetting agent such as a nonionic surfactant (e.g. a polyethoxylated sorbitan ester) is added in preparing this preparation. n COPYRIGHT: (C)1990,JPO |
