abstract PURPOSE: To measure TNF-α with high sensitivity by forming an anti-TNF-α antibody as an Fab' fragment and conjugating enzyme via a maleimide group with the SH group of the hinge part thereof and thereby forming the enzyme antibody complex. n CONSTITUTION: The enzyme is conjugated via the maleimide group with the SH group derived from the disulfide bond in the hinge part of the Fab' fragment of the anti-TNF-α antibody to form the complex. The combination of the complex/antibody which is ≤10 in the relative fluorescent intensity of a zero blank when TNF-α is measured with ELISA by using the above-mentioned complex prepd. from one of the antibodies of two kinds of the different anti- TNF-α as the 2nd antibody and the other antibody as the 1st antibody is used. The quantity of the TNF-α in a soln. (prescribed human bodily fluid, such as blood or urine) is exactly measured with the high sensitivity in such a manner. The measurement of the TNF-α quantity in the normal human serum and the measurement of the fluctuation in the TNF-α values in the diseases arising from various inflammatory symptoms are enabled. n COPYRIGHT: (C)1990,JPO&Japio
