abstract PURPOSE: To easily perform optical resolution by reacting a specific compound with a biocatalyst having asymmetric hydrolysis activity, thereby hydrolyzing one of the optically active isomers. n CONSTITUTION: A biocatalyst such as lipase or immobilized lipase is produced by treating cells of a microbial strain belonging to genus Aspergillus. etc., by extraction, etc. The obtained biocatalyst is added to a water-saturated organic solvent (e.g. benzene) containing 0.5-10wt.% of a racemic propionic acid ester compound of formula [R 1 is acyl (e.g. lower alkanoyl such as acetyl or mono- or di-halogeno-lower alkanoyl such as chloroacetyl); R 2 is ester residue (e.g. lower alkyl such as methyl or ethyl)] and the compound is hydrolyzed at 20-50°C to obtain a reaction liquid. The reaction liquid is treated e.g. by silica gel column chromatography to separate an antipode and obtain an optically active propionic acid ester compound [e.g. optically active threo-3-(4- methoxyphonyl)-3-(2-nitrophenyl-thio)-2-acyloxyproplonic acid ester]. n COPYRIGHT: (C)1990,JPO&Japio
