abstract | PURPOSE: To accomplish assay in a short time with a small amount of enzyme of the title substance for use in acute pancreatitis diagnosis by determining saccharose with fructose-modified oligosaccharide as substrate, produced through glucoamylase or its combination with α-glucosidase. n CONSTITUTION: A fructose-modified oligosaccharide (e.g. isopropylidene matotoheptaosyl frucside) as substrate is made to act on an α-amylase-contg, sample, and an enzyme reagent i.e., glucoamylase or its combination with α- glucosidase is then allowed to act on the resultant oligosaccharide to form saccharose. Thence, sucrose phosphorylase is made to act on this saccharose to form glucose-1-phosphoric acid and fructose, and this fructose is then allowed to react with NADH in the presence of mannitol dehydrogenase to produce NAD, which is then determined, thus accomplishing the objective assay of α-amylase activity. n COPYRIGHT: (C)1990,JPO&Japio |
