titleOfInvention Production of hepatocyte-proliferation factor
abstract PURPOSE: To provide the subject factor extracted and separated from intercellular substance of spleen tissue and having activity to promote the proliferation of hepatocyte in vitro. n CONSTITUTION: Fresh spleen is extracted from a slaughtered animal such as rat, cattle or pig, placed on a mesh of e.g., stainless steel and crushed to loosen the cell cluster. The cells are washed with a proper amount of a physiological salt solution such as physiological salt solution buffered with phosphoric acid to collect the intercellular substance in a container placed under the mesh. The obtained intercellular substance is centrifuged and a soluble fraction containing the hepatocyte-proliferation promoting factor is obtained as the supernatant liquid. The proliferation factor has the following characteristics. (i) Molecular weight is 40,000 and 150,000 or more. (ii) It is not adsorbed to a heparin column. (iii) The hepatocyte-proliferation promoting activity is inactivated by the heat-treatment at 100°C for 2min. (iv) The hepatocyte-proliferation promoting activity is not inactivated in the presence of trypsin (treated at 34°C for 2hr at a concentration of 0.01%).
