titleOfInvention Separation of cell of fungus
abstract PURPOSE: To surely aggregate and separate waste cell of autodigestion without lowering activity of the aimed enzyme by adding chitosan and a polyacrylic acid-based surfactant to a solution of autodigestion of fungus. n CONSTITUTION: Cells are separated and recovered from a culture solution of a fungus such as Hansenula polymorpha by centrifuging. Then the separated cells are subjected to autodigestion by using an autodigestion agent such as benzene or cyclohexane in a nitrogen gas atmosphere. Then the treated solution after the autodigestion is blended with chitosan of polymer flocculant and a surfactant of polyacrylic acid-base such as sodium polyacrylate. Then the treated solution is allowed to stand, centrifuged and filtered by using bleached cotton cloth to give aggregated and separated waste cells, which are separated and removed to give a filtrate containing the aimed enzyme. Consequently, waste cells in the solution of autodigestion can be removed by a simple method without lowering activity of the aimed enzyme. n COPYRIGHT: (C)1990,JPO&Japio
