abstract | PURPOSE: To obtain the title compound useful as a drug intermediate, etc., readily and in high yield, by using a specific acid catalyst and a specific hydrazone as raw materials, subjecting the raw materials to ring closure under heating the then hydrolyzing with an alkali. n CONSTITUTION: A hydrazone compound shown by formula II (R 1 is H, halogen, alkyl, alkoxy or nitro; R 2 is H or lower alkyl; A is tosyl, trichloromethanesulfonyl or methanesulfonyl) is heated in the presence of an acid catalyst at 5-15pts.wt. based on the compound shown by formula II or polyphosphoric acid or 2-6 equivalents based on the compound shown by formula II of p-toluenesulfonic acid and subjected to ring closure to form a indole compound shown by formula III. Then this compound is hydrolyzed in an alcohol solvent in the presence of an alkali under heating while stirring to give the aimed compound shown by formula I useful for synthesizing eudistomidine A, an indole compound having calmodulin antagonism discovered from marine animals in the adjoining seas of Okinawa Prefecture, a drug compound. n COPYRIGHT: (C)1989,JPO&Japio |
