abstract | PURPOSE: To precisely and inexpensively determining an enzyme in a short time, by determining strength of fluorescence of 4-methylumbelliferone obtained by hydrolyzing a specific substrate with N-acetyl-β-D-glucosaminidase. n CONSTITUTION: 4-Methylumbelliferyl-N-acetyl-β-D-glucosaminido (4MU) is blended with citric acid to give a substrate solution (A) having pH 4.3-4.9. Then an enzyme-inactivated skim milk collected from a normal cow is added to defatted product of a milk affected by mastitis and reacted with a definite amount of N-acetyl-β-glucosaminidase (NAG) to provide a standard solution (B). Then, fluorescence strength (C) of 4-methylumbelliferone obtained by adding NAG to the liquid A and subjecting the liquid A to hydrolysis reaction and fluorescence strength obtained by reacting the liquid A with a specimen sample are subjected to comparative determination to measure an enzyme such as NAG of cow milk, etc. n COPYRIGHT: (C)1989,JPO&Japio |
