abstract | PURPOSE: To separate and to purify a large amount of the title superoxide dismutase and catalase simply and in high purity, by treating an hemolyzed solution with an anion exchanger equilibrated with a phosphoric acid buffer solution at a given pH. n CONSTITUTION: A hemolyzed solution of human, etc. is treated with an anion exchanger equilibrated with a phosphoric acid buffer solution at pH 7.2W7.5 and both Cu, Zn type superoxide dismutase (SOD) and catalase are recovered to give a recovered material (A). Then the component A is concentrated, desalted, SOD and catalase as the same fraction are purified by anion exchange chromatography to give a purified fraction (B). Then the component B is passed through a metallic chelating affinity chromatography resin and separated into SOD fraction (C) and a catalase fraction (D). Then the fraction C is purified in the presence of a thiosulfate by chromatography carrying copper, the fraction D is purified chromatography of hydrophobic mutual action to recover erythrocyte derived Cu, Zn type superoxide dismutase and catalase. n COPYRIGHT: (C)1989,JPO&Japio |
