abstract | PURPOSE: To obtain a sandwich immunoassay requiring only one calibrator by employing an antibody specific to the light chain of IgG thereby eliminating the difference of response due to heavy chain subclass. n CONSTITUTION: A specimen containing monoclonal IgG is brought into contact with a support having at least one of antibody anti-K or anti-λ light chain IgG capturing antibody and the reaction mixture is incubated to form a first reaction mixture. Bound monoclonal IgG is then separated from unbound monoclonal IgG through centrifugal separation or filtration. A support having first complex of captured antibody and monoclonal IgG is then brought into contact with a labeled anti-light chain IgG antibody to form a second immunocomplex. Subsequently, the labeled anti-light chain IgG antibody and second immunocomplex are separated from unbound labeled antibody and the labeled quantity in second complex is measured by a measuring means dependent on the used label. n COPYRIGHT: (C)1989,JPO |
