abstract | PURPOSE: To easily measure NADH with super-high sensitivity by leading NADH amplified by the enzyme cycling method in which a specific enzyme is allowed to act on the substrate such as oxalate to chemiluminescence. n CONSTITUTION: The standard solution of NADH or a sample solution containing NADH is combined with enzymes such as maleic acid dehydrogenase(MDH) and alcohol dehydrogenase(ADH) and substrates such as oxalate and ethanol, then allowed to react at about 25°C for about 10minutes to obtain the reaction product. The product, after the enzymes are deactivated, is combined with an excessive amount of MDH, glutamic acid-oxalacetic acid transaminase and glutamate to form NADH in an amount corresponding to the maleate formed by the above-stated reaction. The solution containing formed NADH is combined with 1-methoxyphenazine methosulfate and isoluminol-microperoxidase solution to emit luminescence and NADH is determined by measuring the intensity and using a calibration curve. n COPYRIGHT: (C)1989,JPO&Japio |
