abstract PURPOSE: To decide whether an objective substance is present more than a threshold quantity by causing a sample to react with a fourth labeled substance after a known quantity of second substance specific to the objective substance to be detected and a third substance exhibiting reactivity identical to that of the objective substance are brought into reaction with the sample. n CONSTITUTION: Pregnanediol-3-glucuronide (P 3 G) is caused to be bonded covalently with gelatin and gelatin-P 3 G is adsorbed to a well of microtiter plate. The well is added with a sample solution and a solution containing a known quantity of anti-P 3 G antibody in PBS before being incubated. The solution is then decanted from the well and the plate is cleaned. Subsequently, it is added with a solution containing anti-P 3 G/horseradish peroxidase bond, incubated and cleaned. It is further added with a solution of coloring substrate containing tetramethylbenzidine and hydrogen peroxide and the intensity of coloring is measured. A definite coloring takes place when a specified quantity or more of P 3 G is present in the sample. n COPYRIGHT: (C)1989,JPO
