abstract | NEW MATERIAL:A monoclonal anti-idiotypic antibody against a monoclonal antibody which specifically reacts with rat bladder cancer and human bladder cancer. USE:Preventive, therapeutic agent and diagnostic for bladder cancers. PREPARATION:For example, N-butyl-N-(4-hydroxybutyl)nitrosoamine is given to a rat to generate bladder cancer and the cancer tissues are a rat of the same system to cause immunization. After final immunization, spleen cells are collected. The spleen cells are fused with mouse myeloma cells and cultured in the HAT culture to prepare a hybridoma. The hybridoma is cloned according the limit dilution method to obtain monoclonal hybridoma. Then, the monoclonal hybridoma is given peritoneally to a mouse, dehydrated to give a monoclonal antibody solution. Further, the solution is given to a rat together with adjuvant to effect immunization and cell-fused with spleen cells in the similar manner to effect cloning. The cloned hybridoma is cultured to give anti-idiotypic type antibody. |
