titleOfInvention Sterilization of composition having limited stability
abstract PURPOSE: To sterilize a material compounded with a thermolabile agent under a physiological pH condition by using a buffer solution of a weak acid providing a physiological pH and lowering the pH with a carbonic acid salt and to safely sterilize the above agent at a high temperature by removing carbon dioxide from the above mixture to return the pH of the system to the physiological pH. n CONSTITUTION: A thermolabile pH-sensitive composition in an aqueous mixture is easily and effectively sterilized by preparing a mixture containing (A) the above composition, (B) an aqueous medium buffered to a pH of ≥5.5 or thereabout with a buffer solution other than an amine nitrogen buffer solution, preferably an organic carboxylate buffer solution (especially citrate buffer solution) having a concentration of about ≤50mM and (C) CO 2 of an amount sufficient to lower the pH to about ≤5.5, preferably to 4-5, sterilizing the obtained mixture under a sterilizing condition, preferably at about 121°C for 20min and removing the CO 2 from the mixture to equilibrate the pH of the mixture to a pH of >5.5, preferably to 6-7. n COPYRIGHT: (C)1989,JPO
