abstract PURPOSE: To obtain a sufficiently large pH change and to enable measurement with a high speed by using an antigen or antibody labeled with at least two kinds of enzymes; glucose oxidase and gluconolactonase. n CONSTITUTION: Antihuman sheep IgG labeled by the glucose oxidase and gluconolactonase is first obtd. in the case of detecting, for example, human immune globulin G (human IgG). An ion sensitive (S) FET 1 immobilized with the antihuman sheep IgG on the surface is then immersed for 5min in a soln. 2 to be measured contg. the human IgG to effect an antigen-antibody reaction. Further, this ISFET 1 is immersed for 5min in an antihuman sheep IgG soln. 3 labeled by the glucose oxidase and gluconolactonase. The ISFET 1 is thereafter immersed in a phosphoric acid buffer soln. 4 of 500mg/dl glucose concn., 0.01mM and 6.8pH and the pH change of the film immobilized on the surface of the ISFET 1 is measured.
