abstract | PURPOSE: To readily produce a biocatalyst useful for racemic resolution, etc., of an optically active substance at a low cost by mixing a specific carrier with a lipase and an aqueous buffer solution containing an alcohol, etc., filtering the resultant mixture and then washing the prepared filter cake. n CONSTITUTION: (A) One or more 4-6C N-vinyllactams are reacted with a cyclic acid amide having ≥2 ethylenically unsaturated copolymerizable groups (one or more thereof are directed bound to amide nitrogen atom) to provide an insoluble hardly swelling carrier of a powdery copolymer having 1 μm to 1 mm particle diameter. (B) The extraction treatment of swine pancreas is then carried out to afford a swine pancreatic lipase. (C) One or more 2-6C water-soluble di- to hexahydric alcohols are subsequently dissolved in water to afford a solution. The component A in an amount of 1-50 wt.% is then mixed with 1-30 wt.% component B, 1-68 wt.% component C, 30-97 wt.% water and a pH regulator at pH 5-9 and 0-40°C and the resultant mixture is allowed to stand for 1 min to 3 days to afford a filter cake, which is then washing the produce a biocatalyst. n COPYRIGHT: (C)1989,JPO |
