PURPOSE: To obtain a protective serum, immunizing composition, vaccine, immunizing agent and diagnostic for Manson's blood fluke by using a glycoprotein extracted from hemocyanin of a mollusk such as Megathura clenulata. CONSTITUTION: This agent is composed of a glycoprotein extracted (by proteolysis with pronase) from the hemocyanin (KLH) of a mollask such as Megathura clenulata, having a molar composition of the glucide fraction shown in the table (based on 3 mannose residues) and containing the glucide fraction accounting for 5-5.25% of the molecule. The glycopeptide fraction is obtained by extracting KLH by proteolysis, hydrolyzing the extract and purifying the hydrolyzate by gel-filtration chromatography and has a glucide composition composed of 3 mannose, 4 galactose, 1.6 fucose, 4 glucose, 0.23 xylose, 3.3 N- acetylglucosamine and 2 N-acetylgalactosamine. The glycan fraction is produced by the alkali treatment of KLH or its glycopeptide followed by the N- acetylation and the purification with biogel P-2. |
