abstract | PURPOSE:To obtain a plasmid including autoreproduction of Saccharomycopsis lipolytica, by separating a plasmid from a leucine-demanding Saccharomyces cerevisiae transformed with a gene library. CONSTITUTION:An ampicillin-resistant plasmid is scissored with a restriction enzyme, Leu 2 gene range of Saccharomycopsis lipolytica is isolated, Escherichia coli vector is scissored with a restriction enzyme and connected to the isolated range. The gene library of Saccharomycopsis lipolytica is reproduced by using the connected plasmid. Then leucine-demanding Saccharomycopsis lipolytica as a host is transformed with the gene library. A plasmid is separated from the prepared transformant, Escherichia coli is transformed and the ampicillin- resistant plasmid is separated. |
