abstract | A non-aqueous emulsion method is provided for making polymer microparticles or polymer-coated microparticles. One method is to mix the protein powder and polymer in a hydrocarbon solvent to form a non-aqueous first solution, and add the first solution to the second solution to achieve sustained release. A particulate composition is prepared and the second solution comprises a fluorocarbon liquid and a fluorosurfactant to form a non-aqueous emulsion comprising a plurality of emulsion hydrocarbon droplets in the fluorocarbon liquid. The subsequent microparticle hardening process involves removing the hydrocarbon solvent from the formed emulsion droplets by evaporating the hydrocarbon under ambient conditions with stirring or via vacuum or fluorination. It can be achieved by accelerated curing via the addition of hydrofluoroesters as co-solvents to the carbon. The fluorocarbon liquid is removed and washed with additional fluorocarbon liquid to isolate the sustained release microparticles, which comprise one or more cores of protein and a shell of polymer. |
